Background: Why Fragment HGH?
Human growth hormone (HGH) has complex and sometimes opposing effects — it is anabolic in muscle, lipolytic in adipose tissue, and diabetogenic at high doses. Researchers at Monash University in Australia hypothesized that the lipolytic properties were encoded in a specific region of the HGH molecule. AOD-9604 is the result of isolating that fragment.
What Is AOD-9604?
AOD-9604 corresponds to amino acids 176–191 of the HGH sequence, with a modification at the N-terminus (tyrosine substituted) to improve stability. It was originally developed by Metabolic Pharmaceuticals Ltd as an anti-obesity drug candidate (code: AOD = anti-obesity drug).
Mechanism of Action
AOD-9604 appears to stimulate lipolysis (fat breakdown) and inhibit lipogenesis (fat storage) through mechanisms similar to native HGH, but specifically at the level of adipocytes. Unlike full HGH, it does not appear to stimulate IGF-1 significantly, which is why it avoids the anabolic and diabetogenic effects associated with full HGH administration.
Research Findings
Animal studies demonstrated dose-dependent reduction in body fat in obese rodent models. Human Phase 2 trials showed modest fat reduction but did not meet the efficacy threshold required for Phase 3 advancement as a standalone anti-obesity drug. However, researchers noted benefits in cartilage repair — an unexpected finding that has driven subsequent investigation into AOD-9604 for osteoarthritis applications.
